Abstract

Background: Erkhem-8 medicine is a composed Crocus Sativus L, Lagotis integrifolia, Gentiana barbata froel etc, used for the treatment of respiratory diseases, specially pneumonia and fever or hot disease in Mongolian Traditional Medicine, the objectives of effects of Erkhem-8 medicine to determine the some pharmacological study results of experimental animal model. Aim: To investigate the effect of traditional medicine Erkhem-8 on inflammatory and pain pathology model induced in experimental animals. Materials and Methods: The acute toxicity was studied using the V.B. Prozorovsk method. The anti-inflammatory activity of the Erkhem-8 was evaluated by carrageenan induced paw edema. Wistar albino rats, 220–250 g were divided into four groups with 5 animals for each. The group II, III, IV were treated with 190, 380 and 950 mg/kg of Erkhem-8 orally administered once daily for 5 days before Carrageenan injection. Group I served as control received saline. The rats were then challenged with subcutaneous injection of 0.1 mL of 1% solution of carrageenan into the sub plantar region of right paw. The paw volume was measured before (0 h) and after carrageenan injection at 30, 60, 120, 180, 240 and 300 minut by plethysmometer. Results: The toxicity of the Erkhem-8 medicine was at LD50=9.5 (7.6-11.9) g/kg. The effective dose is ED=190-950 mg/ kg. Erkhem-8 medicine showed a significant decreased in mean paw edema volume in carrageenan induced inflammation. A significant anti oedematous activity at high dose (950 mg/kg) of Erkhem-8 was observed during the 3-6 phases of inflammation. The maximal percentage inhibition was observed at 180, 240 and 300 minutes. In the study of the effect of Erkhem-8 medicine on peripheral pain induced by acetic acid, the number of pain responses was reduced by 42% (p<0.01) in the group treated with Diclodenc 25 mg/kg. Erkhem-8 traditional medicines have the effect of relieving pain of peripheral pain induced by acetic acid, in high doses (p<0.01). Conclusions: These results show that Erkhem-8 medicine has low acute toxicity. In the carrageenan-induced inflammatory disease model, Erkhem-8 medicine at a dose of 950 mg/kg showed an anti-inflammatory effect by reducing the amount of paw edema in chronic inflammation. In peripheral pain caused by acetic acid, Erkhem-8 traditional medicine has analgesic effect at a dose of 950 mg/kg.
