Background: Staphylococcus aureus (S.aureus) is a highly virulence pathogen that responsible for various tissues infection and a wide range of diseases, including food borne infection, skin infections, pneumonia, mastitis and bacteremia in humans and animals. Aim: To compare PCR and LAMP methods in detecting S.aureus. Materials and Methods: In our study, we analyzed a comparison of PCR and LAMP results on S.aureus (ATCC 6538) standard strain used quality control strain solution which diluted, 0.1fg/µL, 1fg/µL, 10fg/µL, 100fg/µL, 1pg/µL, 10pg/ µL, 100pg/µL, 1ng/μL, 10ng/μL, and 100ng/μL DNA concentration from BP agar cultures. A comparison was made using PCR and LAMP methods to detect diluted DNA concentrations. Results: The nuc A gene sensitivity in PCR results is positive in 10fg/µL DNA solution and negative in 0.1fg/µL and 1fg/L DNA. In LAMP test results, it showed a positive response to all concentrations. Conclusion: The LAMP method’s sensitivity is 100 times more than PCR for detecting S.aureus. Therefore this method might be more practical in near future.
