Background: The Philadelphia chromosome is the main cause of chronic myeloid leukemia. The Philadelphia chromosome is conditioned by a chromosomal translocation between chromosomes 9 and 22, creating the BCR-ABL1 gene. Detection and quantification assay of the BCR-ABL1 gene is essential to determine the diagnosis of chronic myeloid leukemia, and to determine drug dosage and treatment results. Aim: To determine identify the Philadelphia chromosome by real time polymerase chain reaction assay in individuals with leukemia, identify variants, and compare treatment outcomes. Materials and Methods: The study was carried out between in August 2022 and December 2022, quantitatively and qualitatively assay evaluated by multiplex and real-time polymerase chain reaction analysis on a total of 15 patient samples. Results: The average age of 15 participants aged 14-72 was 42.9 years. P210 mutation was found in 13 patients during the mutation analysis of the total study participants. the results of treatment and analysis, 13 patients were tested for mutation counting and evaluated by the international standard metrology method. In 6 patients, BCR-ABL1 mutation was highly counted, while in the remaining 7 patients, BCR-ABL1 mutation was at the control level or molecular level and Complete Cytogenetic Response. Conclusion: BCR/ABL1 testing has high therapeutic and diagnostic value in patients with chronic myeloid leukemia.
