A comparative analysis of different nucleic acid extraction methods used to detection of SARS-CoV-2

Background: The reverse-transcription real time-PCR (rRT-PCR) performance has significantly relate with the efÏciency of the viral RNA extraction procedures, yield and purity of nucleic acid and presence of contaminations. In our country, there are various type of nucleic acid extraction methods has applicating for SARS-CoV-2 detection. We hypothesizing the differences of the methods can be the affect as a factor for the result of the test and it`s missing. Aim: The aim of this study was to compare some of the nucleic acid extraction methods used for SARS-CoV-2 detection of clinical laboratories in Ulaanbaatar city. Materials and Methods: Thirteen SARS-CoV-2 positive, one negative nasopharyngeal samples have enrolled in the study. We compared Ct value and qPCR test results of nucleic acid sample, which extracted by five different automated nucleic acid extraction system with the closed-source with their kits. For the statistical analysis, ROC curve analysis, Friedman`s test and Mann Whitney test has performed. Results: The Ct values of amplification on nucleic acid samples extracted by M12 method, was significantly lower comparing with others. According to ROC curve analysis, under area the curve (AUC) were significantly higher for the M12 (AUC=1.00; 95% CI, 0.968-1.00) and S48 (AUC=0.991; 95% CI, 0.951-1.00). Conclusion: Our finding suggests that the M12 and S48 have high efÏcient with lower Ct value and higher AUC. Also, difference between efÏciency of the nucleic acid extraction methods, can be affect as a factor for the detection of SARSCoV-2.
