Abstract

Background: Many traditional recipes such as Juru-5 have been used for the treatment of diabetes mellitus in traditional medicine. There are five plants in Juru-5, Phyllanthus emblica Linn., Curcuma longa Linn., Berberies aristata DC., Thlaspi arvense L., and Tribulus terristris L. The bioactive compounds of these plants have effects to protect pancreatic beta cells and lower fasting glucose level. Objective: The aim of this study was to determine certain bioactive compounds in Juru-5. Method: We weighed 1.0 g of powdered Juru-5 recipe to prepare extract and compared with standard gallic acid and caffeic acid using thin-layer chromatography to determine the total phenolic compound. Moreover, we compared with standard quercetin, caempherol and rutin using thin-layer chromatography to determine the total flavonoids. Results: We compared the bioactive compounds in the traditional Juru-5 recipe with standard substances by thin-layer chromatography. According to our experimental result, phenolic compound (gallic acid Rf=0.95) and its derivatives flavonoids (rutin Rf=0.77, quercetin Rf=0.57, campherol Rf=0.73), respectively, detected. Quantitative concentrations of these bioactive compounds were determined by UV spectrophotometry. Conclusions: The total polypenolic content was 8.31±0.8% (gallic acid equivalents). And the total amount of rutin was determined to be 3.19±0.2% and quercetin was 0.485±0.005% in Juru-5 recipe.
