Abstract

Background: 16S rRNA amplicon sequencing is an accurate method of detecting microbial infection without culture. It is unclear how Helicobacter pylori (H. pylori) infection interacts with other gastric microbiome according to disease status Methods: We enrolled Mongolian patients with the age of 18 who having with dyspepsia in the study. Gastric disease status was evaluated based on histological evaluation namely updated Sydney system score. Results: We examined 161 individuals with a mean age of 43.6 years, and 64.6% were women. H. pylori is the most dominant bacterial species in the stomach. Using diagnostic methods 122 (75.8%) participants were H. pylori positive, the cutoff values were 1113 (real count number) and 4.4% (relative abundance) respectively. By the disease H. pylori count numbers were lowest in the normal group (10823±36215) and highest in the gastric atrophy group (75664±57999) and non-atrophy group (58614±60925) (p<0.01), lower in the intestinal metaplasia group (38007±46131) compared with normal group (p<0.001). The bacterial diversity was significantly highest in the metaplastic atrophy group and the least was in gastritis subgroups. Conclusion: We determined the quantitative number of H. pylori infection and showed how it communicate with other microbiota in different kind of stomach diseases based on 16S rRNA amplicon sequencing approach. We also determined the candidate bacterial species in gastric cancer precursor disease based on gastric microbiome study.
