Determination of peripheral blood lymphocyte count in healthy population
Keywords:
Immunophenotyping, flow cytometry, lymphocyte reference valueAbstract
Background and aim: The mean value of peripheral blood lymphocyte subset is important in diagnosis of immune mediated diseases, clinical observation and prevention of outcomes. There are few reports content of lymphocyte numbers in healthy Mongolian populations. The definition of mean values in the healthy Mongolian blood donors could be helpful also to better define the disease status. This study was aimed to determine the mean values of peripheral blood lymphocyte subsets in blood donors and assess the influence of some factors on cell numbers Materials and methods: Total 255 blood donors who visited the National Center for Transfusion Medicine were recruited. Peripheral blood B (CD3CD19+), T (CD3+ CD19), CD4 T (CD3+ CD4+) and CD8 T (CD3+ CD8+) cell count (cells/µl) was performed by flowcytometry (MACSQuant® analyzer 10) using fluorescent monoclonal antibodies. Results and conclusion: Total 255 blood donors of Mongolian nationality (139 males and 116 females) aged 20-67 year (34.8±11.4) were investigated for determination of peripheral blood lymphocytes. The mean values of the lymphocyte counts were as follows: B cells 163.7 cells/µl (95% Cl, 165.4-186.9), T cells 1000.9 cells/µl (95% Cl, 976.7-1056.9), CD4 T cells 550.3 cells/µl (95% Cl, 540.7-587.3) and CD8 T cells 360.1 cells/µl (95% Cl, 366.8-407.3). Additionally, the CD4/ CD8 ratio was 1.5 (95% Cl, 1.5-1.6). In comparison with age, sex, BMI and blood group mean value was statistically different only for sex. Significantly elevated mean value for B cells (p=0.016), CD4T cells (p=0.002) and CD4/CD8 (p=0.002) were found in female donors comparing with males. Mean value and 95% confidence interval of peripheral blood lymphocytes were established in blood donors. Significantly elevated mean value for B cells, CD4 T cells and CD4/CD8 were found in female donors comparing with males (p<0.05). However there was no significant differences or correlations according to age, BMI and blood types.
