Abstract

Background: Cordiver is a Mongolian medicine compound preparation written by traditional medical source book “Important principles Food and Drink” for the treatment of depression. Therefore, in our present study, we aimed to investigate the antidepressant-like effect of “Cordiver’ preparation in CUMS (chronic, unpredictable, mild, stress) -induced rats. Methods: Rats were divided into four groups (n=10). Group I (control), group II (CUMS). Group III (Cordiver 700mg/ kg+CUMS) and Group IV (neurovalin 343 mg/kg+CUMS). The acute toxicity was studied using the V.B. Prozorovsk method. The CUMS procedure was performed as described previously with minor modifications (Zhang, Junxia Luo et all 2014). Data are expressed mean±SEM (n=10). Level of significance was set at p<0.05. Results: The toxicity of the extracts Cordiver traditional medicine was at LD50=35.5(31-40) g//kg. The effective dose is ED=700 (355-1420) mg/kg. Blood plasma levels of cortisol in the animals of the experimental group were reduced by a statistically significant difference (p<0.05) compared with the control group and increased serotonin levels with statistically significant differences (p<0.05). In addition, the depression and anxiety levels of the experimental animals were reduced by a statistically significant difference (p<0.01) and the movements activity was increased by a statistically significant difference (p<0.05). Conclusions: These results show that Cordiver preparation has low acute toxicity. Cordiver preparation is effective in preventing depressive-like behavior in this CUMS-induced depression model.
