Abstract

Background: 10-15% of the total outpatient of the National Center of Dermatology are children and youths with skin acne. One of the most common skin disorders is acne, which is an inflammatory disease of the sebaceous glands. It is estimated 85% of teenagers and negatively affects the patient’s self-confidence and social life, eventually leads to deep depression. Aim: To prepare anti-acne gel from the medicinal plant (Syzygium aromaticum L.). Materials and methods: The technological parameters of medicinal plant Syzygium aromaticum L plant, liquid extract was prepared by intensive maceration method, and in order to determine the appropriate liquid extract, distilled water, 20%, 40%, and 70% ethanol were taken from the liquid extract, respectively, and selected by comparing the content of polyphenolic compounds in the prepared extract. The content of total polyphenolic compounds Syzygium aromaticum L. extract was determined by spectrophotometrically Folin-Ciocalteu method, transferred to gallic acid. For choosing the appropriate ratio of gel excipients, 3 models were prepared and gels were prepared by dispersion method from liquid extract. The quality of the gel was determined by parameters such as appearance, pH, content of active substances, and quality of application. Antibacterial activity of gel containing Syzygium aromaticum L. was determined by perpendicular inoculation method using 6 standard strains of pathogenic bacteria. Results: When particle size of medicinal plant Syzygium aromaticum L. was 0.85 mm, the highest content of extractive matter was determined 31.061±1.318%, so the appropriate size of raw material was 0.85 mm. It was determined that the internal absorption coefÏcient of plants was 3.08±0.257 at 0.85mm and 2.9±0.296 at 1mm, and the total absorption coefÏcient was 3.934±0.675 at 0.85mm and 3.97±0.675 at 1mm. The extractive matter of plant raw materials were 31.061±1.318% in distilled water, 28.401±1.094% in 20% ethanol, 29.765±1.624% in 40% ethanol and 30.965±1.314% in 70% ethanol. The amount of polyphenolic compounds in the liquid extract(distilled water) prepared by intensive maceration was 0.538±0.0114%. For determining the antibacterial activity of gel containing Syzygium aromaticum L., was active against standard strains of E. faecalis, S. aureus, E. coli, and B. cereus. Conclusion: It was suitable to preparing of liquid extract of Syzygium aromaticum L., the raw material of the plant was cut into 0.85 mm size and extracted by intensive maceration with distilled water as the extragent. The liquid extract had dark brown color, bitter taste, aromatic smell and polyphenolic compound content was 0.538±0.0114%. For preparing gel, it was suitable using 30% of liquid extract of Syzygium aromaticum L. as active ingredient and 1% carbomer as a gelling agent, 5% propylene glycol and 1.5% glycerin as a permeability enhancer and 0.5% menthol to promote antimicrobial activity.
