Abstract

Background: Allergy is hypersensitivity reaction caused by immune mechanisms. Allergic diseases are increasing annually among the world’s population, and bronchial asthma is the most common of allergic lung diseases. According to a 2019 study by the World Health Organization (WHO), there were 339 million cases of allergic bronchial asthma worldwide, and it is estimated that the number will reach 400 million in 2025. Clinical trials are the best way to study allergic asthma, but due to the limitations of clinical trials and ethical issues, researchers use animal models as a alternative to study asthma. Clinical trials are the best way to study allergic asthma, but animal models are often used because of ethical concerns. The need for experimental animal disease models to understand the pathogenesis of allergies, to test new treatments, and to thoroughly study prevention methods is the basis of this research. Aim: To confirm the allergy model induced by ovalbumin and aluminum hydroxide in C57BL/6 mice by histological analysis. Materials and Methods: In this study, 10 female C57BL/6 mice aged 6-8 weeks were divided into control and experimental groups to create an allergy model using a mixture of ovalbumin and aluminum hydroxide. 24 hours after the last dose of ovalbumin, experimental mice were euthanized, and lung histology was performed. The lung tissue specimens were stained with hematoxylin eosin and PAS staining methods to evaluate inflammatory cell infiltration and the number of goblet cells by histological score. Results: Hematoxylin and eosin staining analysis of lung tissue specimens revealed inflammatory cell infiltration, especially eosinophils infiltration, in the pulmonary perivascular and peribronchial region in experimental mice treated with ovalbumin. The lung tissue was stained with (periodic acid Schiff ) PAS staining and analyzed by light microscopy, the number of goblet cells increased (*p<0.01 compared to control group ) and mucus secretion increased in the ovalbumin-treated experimental group compared to the control group. Conclusion: A combination of ovalbumin and aluminum hydroxide induced an allergic lung inflammation model in C57BL/6 mice and was confirmed by histological analysis.
